=== High-performance === The operating principle of CCC equipment requires a column consisting of a tube coiled around a bobbin. The bobbin is rotated in a double-axis gyratory motion (a cardioid), which causes a variable g-force to act on the column during each rotation. This motion causes the column to see one partitioning step per revolution and components of the sample separate in the column due to their partitioning coefficient between the two immiscible liquid phases. "High-performance" countercurrent chromatography (HPCCC) works in much the same way as HSCCC. A seven-year research and development process produced HPCCC instruments that generated 240 g's, compared to the 80 g's of the HSCCC machines. This increase in g-force and larger bore of the column has enabled a ten-fold increase in throughput, due to improved mobile phase flow rates and a higher stationary phase retention. Countercurrent chromatography is a preparative liquid chromatography technique, however with the advent of the higher-g HPCCC instruments it is now possible to operate instruments with sample loadings as low as a few milligrams, whereas in the past hundreds of milligrams had been necessary. == Regulation == Post-translational modifications of prolidase regulate its enzymatic abilities. Phosphorylation of prolidase has been shown to increase its activity while dephosphorylation leads to a decrease in enzyme activity. Analysis of known consensus sequence required for serine/threonine phosphorylation revealed that prolidase contains at least three potential sites for serine/threonine phosphorylation. Nitric oxide, both exogenously acquired and endogenously generated, was shown to increase prolidase activity in a time- and dose-dependent manner via phosphorylation at these serine and threonine sites. == Prognosis == Necrobiosis Lipoidica is classically chronic, difficult to manage, and treat. The prognosis is not reassuring. Squamous cell carcinoma is often a concern with NL. However, this occurs very late into the diagnosis Treatment can typically stop the growth of lesions and ulcers. Lesion treatment is often exquisitely painful and requires intensive wound care. Aesthetically, wounds rarely heal without scarring.
In January 2019, it was reported that some automated aspects of its production were implemented at Oak Ridge National Laboratory in Tennessee, that are expected to triple the number of plutonium pellets produced each week. The production rate is now expected to increase from 80 pellets per week to about 275 pellets per week, for a total production of about 400 grams per year.
