The short version of N-acetyl Semax fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2025-09-14. Anything still debated is marked as such rather than presented as settled.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.
The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.
Terminology around the compound varies by source. It appears in catalogues and papers as Semax, as the heptapeptide ACTH(4-7)-Pro-Gly-Pro, and under various alphanumeric laboratory codes used by individual suppliers. These names refer to the same sequence but may imply different salt forms, purity grades, or counter-ions. Peptide databases usually list the free base mass, while product descriptions sometimes report acetate or trifluoroacetate salts with a different formula weight. Because naming conventions for research peptides are not standardised across vendors, checking the declared sequence and measured mass is more reliable than relying on a trade name alone.
Identity and purity are commonly assessed by reversed-phase high-performance liquid chromatography, with ultraviolet detection near 214 nanometers for the peptide backbone. Mass spectrometry, either electrospray or matrix-assisted laser desorption, confirms molecular mass and detects truncation or modification products. Amino acid analysis can verify composition. Because the sequence contains no strongly absorbing aromatic residue apart from phenylalanine, detection wavelengths are chosen carefully. Purity values above 95 percent are typical for research-grade material.
Quality documentation for research-grade peptide usually includes a certificate of analysis stating purity, identity, and residual counterion content. Batch-to-batch variation in purity and salt form is a recognized issue, and comparisons across suppliers require attention to the exact counterion, for example acetate versus trifluoroacetate. Sequence verification by tandem mass spectrometry provides stronger evidence than a single mass measurement. For any study reporting biological results, the analytical method and the measured purity should be stated explicitly. Independent verification of supplier claims is considered good practice.

Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.

== History == The company was founded in April 2012 by former Morgan Stanley investment banker Adam Forste, who formed Bay Grove Capital, LLC, through the consolidation of acquired warehousing and logistics companies beginning, in December 2008, with the purchase of Seafreeze from Toyo Susian Kaisha. The company then acquired CityIce, in 2009, and Flint River Services, in 2010, followed, in 2011, by Terminal Freezers of Santa Maria, California, and Richmond Cold Storage, purchased from Atlanta Equity. Lineage was formed with the fifth-largest network of temperature-controlled warehouse facilities in North America in 2012, according to International Association of Refrigerated Warehouses (IARW) rankings, with 40 facilities in eight states, including near international ports at Savannah, Georgia, Seattle, Washington and Norfolk, Virginia; as well as providing warehouse management, consulting, and food processing support. In a March 2021 interview with Bloomberg, Lineage CEO Greg Lehmkuhl said the company is actively preparing its initial public offering (IPO). The company completed its IPO in July 2024 with a listing on the Nasdaq, raising around $4.5 billion, valuing the company at over $18 billion. On April 21, 2024, a fire broke out at the Lineage cold storage facility in Benton County, Washington. The fire burned for 60 days. On June 17, 2026 a large fire broke out at the Lineage cold storage facility in Boyle Heights, Los Angeles, accelerated by the large facility's solar panels encompassing its entire roof, refrigerants, and decaying food within the building.
Throughout the process, it is necessary to keep close control of the moisture content using a moisture meter system in order to reduce over-drying and allow operators to know when to pull the charge. Preferably, this in-kiln moisture meter will have an auto-shutoff feature.

== Manufacture == Blotting paper is made from different materials of varying thickness, softness, etc. depending on the application. It is often made of cotton and manufactured on special paper machines. Blotting paper is reputed to be first referred to in the English language in the 15th century but there is a tradition in Norfolk, England that it was invented by accident at Lyng Mill on the River Wensum. It is reported that a Berkshire (England) paper mill worker failed to add sizing to a batch of paper that was being produced. The batch was discarded. Subsequently, someone tried to write on a piece of this discarded "scrap" paper and found that it rapidly absorbed any ink applied, making it unusable for writing. Its marked absorbency having been noted, however, led to its subsequently being produced and used as blotting paper, replacing sand, which was the material that had been used for absorbing superficial wet ink. In a time when most paper was produced from "rags", red/pink rags, from which it was difficult to remove all colour and had generally been discarded, were now directed to the production of blotters, hence the historically characteristic pink colour of blotters.
Chemical specificity is the ability of binding site of a macromolecule (such as a protein) to bind specific ligands. The fewer ligands a protein can bind, the greater its specificity. Specificity describes the strength of binding between a given protein and ligand. This relationship can be described by a dissociation constant, which characterizes the balance between bound and unbound states for the protein-ligand system. In the context of a single enzyme and a pair of binding molecules, the two ligands can be compared as stronger or weaker ligands (for the enzyme) on the basis of their dissociation constants. (A lower value corresponds to a stronger binding.) Specificity for a set of ligands is unrelated to the ability of an enzyme to catalyze a given reaction, with the ligand as a substrate. If a given enzyme has a high chemical specificity, this means that the set of ligands to which it binds is limited, such that neither binding events nor catalysis can occur at an appreciable rate with additional molecules. An example of a protein-ligand pair whose binding activity can be highly specific is the antibody-antigen system. Affinity maturation typically leads to highly specific interactions, whereas naive antibodies are promiscuous and bind a larger number of ligands. Conversely, an example of a protein-ligand system that can bind substrates and catalyze multiple reactions effectively is the Cytochrome P450 system, which can be considered a promiscuous enzyme due to its broad specificity for multiple ligands.
==== Glycol chitosan ==== Enzymatic activity of intact and PEGylated lysozyme can be evaluated using glycol chitosan by reacting 1 mL of 0.05% (w/v) glycol chitosan in 100 mM of pH 5.5 acetate buffer and 100 μL of the intact or PEGylated protein at 40 °C for 30 min and subsequently adding 2 mL of 0.5 M sodium carbonate with 1 μg of potassium ferricyanide. The mixture is immediately heated, boiled for 15 minutes, and cooled for spectral analysis at 420 nm. As the enzymatic activity to hydrolyze β-1,4- N-acetylglucosamine linkage was retained after PEGylation, there was no decay in the enzymatic activity by increasing the degree of PEGylation.
According to Richards, Campania, & Muse-Burke, "There is growing evidence that is showing emotional abilities are associated with pro-social behaviors such as stress management and physical health." Their research also concluded that people who lack emotional expression are inclined to anti-social behaviors (e.g., substance use disorder and alcohol use disorder, physical fights, vandalism), which reflects one's mental health and suppressed emotions. Adults and children who face mental illness may experience social stigma, which can exacerbate the issues.

It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
It contains the 4-7 fragment of adrenocorticotropic hormone extended at the C terminus. That truncation removes the region associated with steroidogenic activity. The design intent was to keep neurotrophic properties while avoiding full hormonal effects.
No natural source of the intact heptapeptide is known. It is produced by solid-phase peptide synthesis. Some related ACTH fragments arise as hormone processing products, but the Pro-Gly-Pro extension is a designed addition rather than a natural sequence.
Solid peptide is best kept frozen at about minus 20 degrees Celsius in a sealed container with desiccant. It should be allowed to reach room temperature before opening to prevent condensation. Repeated warming and cooling of the same vial is discouraged.
