This is a working overview of reversed-phase HPLC, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-12-24. Anything still debated is marked as such rather than presented as settled.
Identity and purity are confirmed with reversed-phase high-performance liquid chromatography, typically monitored at 214 nanometres where the peptide bond absorbs. Mass spectrometry, either electrospray or MALDI-TOF, verifies molecular mass against the theoretical value and detects truncation or adduct formation. Amino acid analysis and peptide mapping provide additional confirmation when required. The most frequently reported impurities are deletion sequences from incomplete coupling, methionine sulfoxide from oxidation, and dimeric species formed through non-covalent aggregation. Impurity profiles depend strongly on the synthesis and purification route chosen by the producer.
Published research has focused mainly on neurological and cognitive endpoints in animal models, with proposed mechanisms involving brain-derived neurotrophic factor and related signalling pathways. A substantial share of the human data originates from a limited number of research groups, and independent replication in other countries remains sparse. Regulatory status reflects that distribution: the peptide is registered as a medicine in Russia and appears in some neighbouring markets, while elsewhere it is handled as a research chemical without approved therapeutic labelling. Questions about dose-response relationships, long-term effects, and comparability across studies are still open.
Lyophilised powder is normally kept at -20 °C in a desiccated container, with some suppliers recommending -80 °C for long-term archival storage. Repeated freeze-thaw cycles are the most common cause of avoidable loss, so aliquoting before freezing reduces variability between working sessions. Dissolved peptide is far less stable than the dry solid and is usually prepared fresh or held briefly at 4 °C. Aqueous solutions support both hydrolysis of the backbone and oxidation of the N-terminal methionine, and these two routes dominate degradation under ordinary laboratory conditions.
Common degradation pathways include oxidation of the methionine side chain, hydrolysis of the peptide backbone, and aggregation under unfavourable pH or concentration. Stability studies typically monitor the main peak by chromatography and report total related substances as a percentage. Because no official monograph exists, acceptance criteria vary between laboratories, and reported purity values are not directly comparable across suppliers. Analysts therefore document the method, column, and detection wavelength alongside each result, and open questions remain about how much biological activity the oxidised forms retain.
Solid semax is typically supplied as a lyophilised powder that is hygroscopic and sensitive to moisture, light, and repeated temperature cycling. Long-term storage of the dry peptide is generally recommended at approximately -20 degrees Celsius, while shorter working periods may use refrigeration at 2 to 8 degrees Celsius. Vials should remain tightly closed and desiccated when brought to room temperature, because condensation can damage the material before it is weighed. Dividing a batch into aliquots is preferable to thawing one container repeatedly.
Dissolution is normally performed in water, phosphate-buffered saline, or normal saline, since the peptide is freely soluble in aqueous media and is rarely handled with strong organic solvents. Solution pH should be kept near neutral, because extreme acidity or alkalinity accelerates backbone hydrolysis and encourages oxidation of the methionine side chain. Once dissolved, the material is less stable than the dry powder and is commonly divided into single-use portions and frozen. Buffers containing primary amines, such as Tris, are often avoided because of possible side reactions.

Circulation time for the peptide is short because peptidases cleave it readily. The Pro-Gly-Pro tail is thought to slow breakdown compared with the bare ACTH fragment, but the gain appears modest. Absorption after intranasal dosing is limited, and only a fraction of a dose is expected to reach the central nervous system. Laboratory concentrations therefore sit well above levels achieved systemically, a gap that complicates translation from bench findings to clinical claims.
The mechanisms attributed to semax are inferred from animal and cell studies rather than traced to one confirmed target. The most frequently cited pathway involves increased expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal and cortical tissue. Some work points to engagement of melanocortin receptors, particularly MC4, which the parent ACTH fragment can activate. Effects on dopaminergic and serotonergic signalling have also been reported. No single account explains all observed results, and the relative weight of each pathway remains unsettled.
Semax is supplied as a lyophilized powder that appears white to off-white. It dissolves readily in water, phosphate-buffered saline, and other aqueous media, which simplifies preparation of working solutions for laboratory use. The nasal products registered in Russia are dilute aqueous solutions, typically around 0.1 percent peptide by weight. Organic solvents are rarely necessary and can complicate handling. Because the peptide is hygroscopic, weighing should be performed quickly and with minimal exposure to ambient humidity.
Solid material is normally kept at minus 20 degrees Celsius in a sealed, desiccated container. Reconstituted solutions are less stable and are usually divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are avoided because they promote aggregation and loss of activity. Light exposure is minimized by using amber glassware or foil wrapping. Published stability data for this peptide are sparse, so recommended storage conditions rest mainly on general practice for short synthetic peptides rather than on dedicated study.
Identity and purity are commonly assessed by reversed-phase high-performance liquid chromatography, with ultraviolet detection near 214 nanometers for the peptide backbone. Mass spectrometry, either electrospray or matrix-assisted laser desorption, confirms molecular mass and detects truncation or modification products. Amino acid analysis can verify composition. Because the sequence contains no strongly absorbing aromatic residue apart from phenylalanine, detection wavelengths are chosen carefully. Purity values above 95 percent are typical for research-grade material.
=== Use of the term biology === From Greek βίος (bíos) 'life', (from Proto-Indo-European root *gwei-, to live) and λογία (logia) 'study of'. The compound appears in the title of Volume 3 of Michael Christoph Hanow's Philosophiae naturalis sive physicae dogmaticae: Geologia, biologia, phytologia generalis et dendrologia, published in 1766. The term biology in its modern sense appears to have been introduced independently by Thomas Beddoes (in 1799), Karl Friedrich Burdach (in 1800), Gottfried Reinhold Treviranus (Biologie oder Philosophie der lebenden Natur, 1802) and Jean-Baptiste Lamarck (Hydrogéologie, 1802). Before biology, there were several terms used for the study of animals and plants. Natural history referred to the descriptive aspects of biology, though it also included mineralogy and other non-biological fields; from the Middle Ages through the Renaissance, the unifying framework of natural history was the scala naturae or Great Chain of Being. Natural philosophy and natural theology encompassed the conceptual and metaphysical basis of plant and animal life, dealing with problems of why organisms exist and behave the way they do, though these subjects also included what is now geology, physics, chemistry, and astronomy. Physiology and (botanical) pharmacology were the province of medicine. Botany, Zoology, and (in the case of fossils) Geology replaced natural history and natural philosophy in the 18th and 19th centuries before biology was widely adopted.
== Mechanism of action == The perceived sweetness of aspartame (and other sweet substances like acesulfame potassium) in humans is due to its binding of the heterodimer G protein-coupled receptor formed by the proteins TAS1R2 and TAS1R3. Rodents do not experience aspartame as sweet-tasting, due to differences in their taste receptors.
The Boers struck first on 12 October at the Battle of Kraaipan, an attack that heralded the invasion of the Cape Colony and Natal between October 1899 and January 1900. With speed and surprise, the Boers drove quickly towards the British garrison at Ladysmith and the smaller ones at Mafeking and Kimberley. The quick Boer mobilisation resulted in military successes against scattered British forces. Sir George Stuart White, commanding the British division at Ladysmith, unwisely allowed Major-General Penn Symons to throw a brigade forward to the coal-mining town of Dundee (also reported as Glencoe), surrounded by hills. This became the war's first major clash, the Battle of Talana Hill. Boer guns began shelling the British camp from the summit of Talana Hill at dawn on 20 October. Penn Symons immediately counter-attacked: His infantry drove the Boers from the hill, for the loss of 446 British casualties, including Penn Symons. Another Boer force occupied Elandslaagte, which lay between Ladysmith and Dundee. The British under Major General John French and Colonel Ian Hamilton attacked to clear the line of communications to Dundee. The resulting Battle of Elandslaagte was a clear-cut British tactical victory, but White feared more Boers were about to attack his main position and ordered a chaotic retreat from Elandslaagte, throwing away the advantage gained. The detachment from Dundee was compelled to make an exhausting cross-country retreat to rejoin White's main force. As Boers surrounded Ladysmith and opened fire with siege guns, White ordered a major sortie against them.
== Biological roles == Highly dynamic disordered regions of proteins have been linked to functionally important phenomena such as allosteric regulation and enzyme catalysis. Many disordered proteins have the binding affinity with their receptors regulated by post-translational modification, thus it has been proposed that the flexibility of disordered proteins facilitates the different conformational requirements for binding the modifying enzymes as well as their receptors. Intrinsic disorder is particularly enriched in proteins implicated in cell signaling and transcription, as well as chromatin remodeling functions. Genes that have recently been born de novo tend to have higher disorder. In animals, genes with high disorder are lost at higher rates during evolution.

== Class B peptidomimetics == This class of peptidomimetics encompasses peptides with a large number of non-natural amino acids, major backbone modifications or larger non-natural building fragments that resemble the conformation of a particular peptide binding motif. Examples involve D-peptide and peptidic foldamers such as beta-peptides.
== History == Clomipramine was developed by Geigy as a chlorinated derivative of imipramine. It was first referenced in the literature in 1961 and was patented in 1963. The drug was first approved for medical use in Europe in the treatment of depression in 1970, and was the last of the major TCAs to be marketed. In fact, clomipramine was initially considered to be a "me-too drug" by the FDA, and in relation to this, was declined licensing for depression in the United States. As such, to this day, clomipramine remains the only TCA that is available in the United States that is not approved for the treatment of depression, in spite of the fact that it is a highly effective antidepressant. Clomipramine was eventually approved in the United States for the treatment of OCD in 1989 and became available in 1990. It was the first drug to be investigated and found effective in the treatment of OCD. The benefits in OCD were first reported by Juan José López-Ibor in 1967, and the first double-blind, placebo-controlled clinical trial of clomipramine for OCD was conducted in 1976, with more rigorous clinical studies that solidified its effectiveness conducted in the 1980s. It remained the "gold standard" for the treatment of OCD for many years until the introduction of the SSRIs, which have since largely superseded it due to greatly improved tolerability and safety (although notably not effectiveness).
When SCLC is found with one or more differentiated forms of lung cancer, such as squamous cell carcinoma or adenocarcinoma, the malignant tumor is then diagnosed and classified as a combined small cell lung carcinoma (c-SCLC). Small-cell lung carcinoma can occur in combination with a wide variety of other histological variants of lung cancer, including extremely complex malignant tissue admixtures.
The Defense Secretary threatened Anthropic, the owner of Claude AI (the only AI authorized by the Department of Defense in early 2026), to designate the company a supply chain risk and force the company to eliminate its restrictions on the use of its AI by the Defense Department or he will use the Defense Production Act to compel them. The Defense Department gave Anthropic a deadline of February 27, 2026. The contract Sec. Hegseth wanted to cancel was worth $200 million. A senior defense official stated that the company will "pay a price for forcing our hand like this." Anthropic stated that they wanted assurances that the DoD would not use their AI tools to spy on Americans or develop weapons that could fire without human involvement. The Defense Department also stated in February 2026 that Grok, OpenAI and Google were close to being approved for use. Claude was used by the Pentagon during their January 2026 operation to capture Nicolas Maduro. In May 2026, the Defense Department announced AI use deals with SpaceX, OpenAI, Google, NVIDIA, Reflection, Microsoft and Amazon Web Services. On February 27, 2026, Hegseth used the Federal Acquisition Supply Chain Security Act to declare Anthropic a supply chain risk and excluded it from all federal contracts. President Trump also stated that every federal agency must stop using Anthropic's AI technology. ChatGPT signed a deal with the Defense Department the same day but their CEO Sam Altman said, "the AI system shall not be intentionally used for domestic surveillance of U.S.

== History == Oxandrolone was first made by Raphael Pappo and Christopher J. Jung while at Searle Laboratories, now part of Pfizer and they first described the drug in 1962. They were immediately interested in oxandrolone's very weak androgenic effects relative to its anabolic effects. It was introduced as a pharmaceutical drug in the United States in 1964. Oxandrolone was prescribed to promote muscle regrowth in disorders which cause involuntary weight loss, and is used as part of treatment for HIV/AIDS. It had also been shown to be partially successful in treating cases of osteoporosis. However, in 1989, in part due to bad publicity from its illicit use by bodybuilders, production of Anavar was discontinued by Searle Laboratories. It was picked up by Bio-Technology General Corporation, which changed its name to Savient Pharmaceuticals. In 1995, following successful clinical trials, Savient released it under the brand name Oxandrin. As of 2011 BTG subsequently had won approvals for orphan drug status by the Food and Drug Administration for treating alcoholic hepatitis, Turner syndrome, and HIV-induced weight loss and as an offset to protein catabolism caused by long-term administration of corticosteroids.
Another suggests that StAR acts in conjunction with PBR, causing the movement of Cl− out of the mitochondria to facilitate contact site formation. However, evidence for an interaction between StAR and PBR remains elusive.

Reversed-phase HPLC gives the main purity figure, most often with UV detection near 214 nanometres. Mass spectrometry then confirms the molecular mass. Together the two methods distinguish a correct sequence from a closely related impurity.
The main chemical risks are methionine oxidation and backbone hydrolysis in solution. Moisture and repeated temperature cycling accelerate both processes. Dry powder held cold and desiccated is considerably more stable than any reconstituted preparation.
It holds a medicine registration in Russia, where it has been used clinically for decades. In most other countries it is treated as a research chemical. That split explains the uneven distribution of clinical literature.
Dry powder is usually kept at about -20 degrees Celsius for long-term storage and at 2 to 8 degrees Celsius for short working periods. Containers should stay sealed and protected from light and moisture to limit degradation.
