Kissinger finally offered the concession that the United States would use "maximum influence" to pressure the South Vietnamese government to release all Viet Cong prisoners within sixty days of a peace agreement being signed. Thiệu once again rejected the peace agreement, only to receive an ultimatum from Nixon: "You must decide now whether you desire to continue our alliance or whether you want me to seek a settlement with the enemy which serves U.S. interests alone". Nixon told Kissinger: "Brutality is nothing. You have never seen it if this son-of-a-bitch doesn't go along, believe me". Nixon's threat served its purpose, and Thiệu reluctantly accepted the peace agreement. As expected, deramciclane reaches greatest peak plasma concentrations with intravenous administration, followed by intraperitoneal, then oral administration with the lowest peak plasma concentration. Studies assessing the elimination half-life of deramciclane point to a range of 20–32 hours for T1/2. The elimination half-life appears to increase with dosage. There is some evidence for accumulation of deramciclane, though it is a topic of debate. Deramciclane undergoes side chain modification and oxidation at multiple positions on the molecule. The side chain reaction forms phenylborneol and N-desmethylderamciclane which is the active metabolite of deramciclane. Oxidation of the molecule results in many hydroxy-, carboxy-, and N-oxide derivatives. Clinical studies investigating the effects of food or lack thereof on deramciclane adsorption show that there is a statistically significant, but not clinically relevant, increase in bioavailability of deramciclane when administered with food because the point of critical instability of deramciclane is relatively low at a pH of 2. ===== Exopeptidases ===== Exopeptidases are enzymes that can cleave the end of an amino acid side chain mostly through the addition of water. Exopeptidase enzymes exist in the small intestine. These enzymes have two classes: aminopeptidases are a brush border enzyme and carboxypeptidases which is from the pancreas. Aminopeptidases are enzymes that remove amino acids from the amino terminus of protein. They are present in all lifeforms and are crucial for survival since they do many cellular tasks in order to maintain stability. This form of peptidase is a zinc metalloenzyme and it is inhibited by the transition state analog. This analog is similar to the actual transition state, so it can make the enzyme bind to it instead of the actual transition state, thus preventing substrate binding and decreasing reaction rates. Carboxypeptidases cleave at the carboxyl end of the protein. While they can catabolize proteins, they are more often used in post-transcriptional modifications.
